issue 一般指社会,政治,经济等方面的大问题.
The mayor is challenged with a big economic problem.那位市长面临着一个经济方面的问题.

problem 泛指（数学,物理等）难题或待解决的问题,如：
This problem on the environmental protection floored the new mayor.
这个关于环境保护的问题把新任市长难住了.

question 是有疑问,有质疑,需要弄清楚真相的问题.
