Small aliquots (c.a.5 μl) of a 0.5 M FITC solution,prepared
in anhydrous DMSO,were added every 5 minutes to
5.0 ml of a 23 uM APA V solution,prepared in a carbonate
buffer,pH 9,at 4°C,with constant stirring,to get a 1:1 APA
V:FITC final ratio.The progress of the reaction was followed
by thin layer chromatography (TLC),using a chloroform:
dichloromethane:methanol (2:1:2,volume ratio) mixture
as developing solvent.After 8 h,the reaction was
stopped by adding an NH4Cl solution up to a final concentration
of 50 mM.Then,As+5 was reduced to As+3 with a β-
mercaptoethanol (2 M final concentration) overnight (O.N.)
incubation at 4°C.
